Puerto Madryn bus terminal is on the corner of Hipolito Yrigoyen and San Martin. It is 3 blocks away from Sentir Patagonia Hostel. There bus services to Trelew (1 hr), Comodoro Rivadavia, Rio Gallegos (over night) and Buenos Aires (19 hrs). There is no direct bus to El Calafate. If you want to visit the Moreno Glacier after seeing right whales then you have to catch a bus to Rio Gallegos and then switch to a bus going to El Calafate.

There is an airport in Puerto Madryn and one with more flights 65kms away in Trelew. Both airports have shuttle bus services that will take you directly to your hostel. There are daily flights to Buenos Aires.
